Isolation, Identification, and Metabolic Characteristics of a Quinoline Degrading Bacterium

A bacterial strain, BW004 that utilized quinoline as its sole carbon, nitrogen, and energy source, was isolated from the activated sludge of the coking wastewater treatment plant in Wuhan Iron and Steel Group, Hubei Province, China. The strain was identified as Pseudomonas sp. according to the 16S rRNA sequencing. The optimum degradation conditions determined by response surface methodology (RSM) were pH 7, rotation 180 r/min, and temperature 30.0℃. The degradation experiments revealed that BW004 could degrade 200?1000 mg/L of quinoline over 98.8% within 4?12 hours. During the degradation, quinoline was first transformed to 2-hydroxyquinolin and 2,8-dihydroxyquinolin, and ammonium was released promptly as the final product of heterocyclic nitrogen; then the dual-ring structure was destroyed and all organics were mineralized in 12?24 hours.

The mechanisms, related enzymes, and research hotspots of microbial nitrification and denitrification are discussed. The operation procedure of molecular ecological technique and its application on the analysis of microbial community structure are summarized. This paper also explores the principles, operation processes, advantages and defects, and application on the study of nitrification and denitrification of three techniques (fluorescent in situ hybridization, denaturing gradient gel electrophoresis, and terminal restriction fragment length polymorphism). Previous studies proved that the molecular ecological technique becomes a powerful tool on the study of nitrification and denitrfication process in the environment.

Biological Removal of a Bacterial Strain Pseudomonas sp. BC001 on Pyridine and Quinoline

An aerobic Gram-negative bacterium BC001 was isolated from the activated sludge in the coking wastewater treatment plant of Capital Iron and Steel Corporation and identified as Pseudomonas sp. according to 16S rDNA gene sequence analysis and physiological characters. The strain grew well in the high concentrated substrates of pyridine (about 400mg/L) and quinoline (about 500mg/L). For pyridine, the biological removal was only by biosorption, while for quinoline, it included two phases of biosorption and biodegradation. The strain could utilize quinoline as its sole carbon source and nitrogen source, and the biodegradation would be promoted by external carbon source with proper dosage. Main intermediate products in the metabolic pathway of quinoline were detected as 2(1H)-quinolinone and 8-hydroxycoumarin, and NH4+ was confirmed to be the final nitrogen metabolic product.

Research on Bio-Regeneration Process of the Ammonium Adsorption Capacity of Natural Zeolite

Using the natural clinoptilolite from Jinyun County, Zhejiang Province as the experimental samples, a static bio-regeneration experiment was conducted for the zeolite saturated with ammonium. The ammonium adsorption capacity of the saturated zeolite were recovered 61%~85% after 73 days by biological process and ion exchange. An on-site pilot experiment of bio-regeneration was also conducted for the zeolite absorted large amount ammonium in Dianchi Lake basin, Yunnan Province. The ammonium adsorption capacity of the zeolite were recovered 22%~36% after 4~6 months by natural biological process.

During May to June 2002, ambient total suspended particulates (TSP) were sampled at Dingri (28.655°N, 87.115°E, 4300 m a.s.l.) and at the north valley of Mt. Qomolangma (28.193°N, 86.827°E, 5000 m a.s.l.). Be-7 radioactive concentrations of the TSP samples were between 0.76~1.7 mBq·m-3STP. The results showed no evidence of contributions from stratosphere or upper troposphere transport to the high ozone concentrations on the surface during the observation period.

The Hourly Measurement of Aerosol and Related Gases in Guangzhou

The measurement was made in July and November 2000 in Guangzhou. The measurement species included SO42-,NO3-,NO2-,Cl- of aerosol and related gases. The hourly variation of the concentration was presented. It showed that NO3-,NO2-,Cl- of aerosol and HCl, HNO2,HNO3 of gas varied with regularity. The variation of gas SO2 and aerosol SO42- depended on the emission of SO2. The ratio of Sa/St was much higher in November than July.

Establishment and Study of the Gas Phase Calibration Source of Hydroxyl Radical

The gas phase calibration source of hydroxyl radical was established using photolysis of H2O at 184.9 nm UV-light produced by low-pressure Hg lamp. The concentration of OH was inferred from the concentrations of O3 and H2O in the flow. The OH concentration was also measured using salicylic acid liquid phase scrubbing and high performance liquid chromatography (HPLC) technique. The relationship between the calculated and measured OH concentrations was relatively uniform. The surface effect in the photolysis tube was studied using laser-induced fluorescence signal and was not of importance.

An Experimental Study for Atmospheric Chemistry of soprene with OH Radicals in the Presence of NOx

The experimental simulation was employed to study the atmospheric chemistry of isoprene with OH radicals in the presence of NOx, using long-path Fourier transform infrared (LP-FTIR) method. OH radical was produced from photodissociation of CH3ONO under irradiation of black lights. Results showed that carbonyl compounds, organic nitrates, HCOOH and CO, were formed. Methacrolein (CH2=C(CH3)CHO), methyl vinyl ketone (CH3C(O)CH=CH2) and HCHO were identified. At the temperature of 298K and the pressure of 93.3kPa, yields of methacrolein and methyl vinyl ketone were (20.8±1.5)% and (33.4±2.2)% respectively. The pressure variation did not influence their yields. Finally the reaction of isoprene with OH radical in the presence of NOx was discussed briefly.

The study of the determination method of dimethyl sulfide (DMS)in sea water is presented in the article. Sample treatment,sample storage, the cold trap temperature and the gas chromatography analysis method is discussed. The detection limit is 0.5 ng DMS. Precision is 7%. Concentrations of DMS in sea water near Qingdao in March, May, and October 1996 are also presented.

The Study of Urban Photochemical Smog Pollution in China

Based on the studies on photochemical smog in Xigu and Yanshan petroleum industrial areas, Beijing city and Guangzhou City, the current pollution level of photochemical smog and its characteristics in China are discussed, its formation mechanism is explored, and its changing tendency in near future is predicted. The results show that photochemical smog pollution is serious in above areas and the effective way to control the photochemical smog in China is to decrease NOx emission. The ozone peak appears not only earlier, but also is higher year by year at Zhongguancun, Beijing, which phenomenon indicates that the atmospheric photochemical activity is enhanced. As the fast increasing of the vehicle population, the air pollution in some big cities is in a transition from coal burning caused problem to automobile exhaust related pollution or the pollution combined them.
